NM_000302.4(PLOD1):c.1027G>A (p.Glu343Lys) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E343K variant (also known as c.1027G>A), located in coding exon 10 of the PLOD1 gene, results from a G to A substitution at nucleotide position 1027. The glutamic acid at codon 343 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
