Pathogenic for Tay-Sachs disease — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000015.10:g.(?_72374460)_(72375992_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 1 of the HEXA gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 1 of the HEXA gene. This is expected to result in an absent or disrupted protein product. A 7.6kb deletion of the promoter and exon 1 has been reported in several French Canadian individuals affected with Tay-Sachs disease (PMID: 3754980). Loss-of-function variants in HEXA are known to be pathogenic (PMID: 1833974, 8490625). For these reasons, this variant has been classified as Pathogenic.
