Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000006.12:g.(?_162201121)_(162443487_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 2-4 of the PRKN gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Similar deletions of exons 2-4 have been observed in individuals and families affected with autosomal recessive early-onset Parkinson's disease and in an individual with clinical features of Parkinson's disease with Lewy bodies (PMID: 10824074, 10939576, 23401296, 23880019). Loss-of-function variants in PRKN are known to be pathogenic (PMID: 10072423, 20301651, 22956510). For these reasons, this variant has been classified as Pathogenic.
