Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.10:g.(?_132557325)_(132557463_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 1 of the RAD50 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 1 of the RAD50 gene. This is expected to result in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with RAD50-related conditions. Loss-of-function variants in RAD50 are known to be pathogenic (PMID: 16385572, 19409520). For these reasons, this variant has been classified as Pathogenic.
