Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.11:g.(?_43099755)_(43125483_?)del, citing Invitae Variant Classification Sherloc (09022015): Similar deletions have been observed in individuals with clinical features of hereditary breast and ovarian cancer (PMID: 18704682, 29433453). This variant is a gross deletion of the genomic region encompassing exons 1-7 of the BRCA1 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 7 of the BRCA1 gene. This is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA1 are known to be pathogenic (PMID: 20104584). For these reasons, this variant has been classified as Pathogenic.
