Likely pathogenic for Juvenile polyposis syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000010.11:g.(?_86912230)_(86919479_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an in-frame deletion of the genomic region encompassing exons 8-10 of the BMPR1A gene. It preserves the integrity of the reading frame. This variant has not been reported in the literature in individuals with BMPR1A-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This variant disrupts a large portion of the cytoplasmic and protein kinase domains of the BMPR1A protein, which are required for the regulation of the TGF- /BMP pathways (PMID: 8397373, 14526373, 23433720). While functional studies have not been performed to directly test the effect of this variant on BMPR1A protein function, this suggests that disruption of this region of the protein is causative of disease. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively.
