This variant would be expected to be in-frame, preserving the integrity of the reading frame. This variant has not been reported in the literature in individuals affected with NBN-related conditions. This variant disrupts the MRE11 interaction domain of the NBN protein, which has been shown to be essential for viability and tumor suppression (PMID: 28076792). While functional studies have not been performed to directly test the effect of this variant on NBN protein function, this suggests that disruption of this region of the protein is causative of disease.
