Pathogenic for Pyridoxine-dependent epilepsy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000005.10:g.(?_126582831)_(126584032_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with ALDH7A1-related conditions. For these reasons, this variant has been classified as Pathogenic. This variant is a gross deletion of the genomic region encompassing exon(s) 4-5 of the ALDH7A1 gene. This deletion is out-of-frame, and is expected to create a premature termination codon and result in an absent or disrupted protein product. Loss-of-function variants in ALDH7A1 are known to be pathogenic (PMID: 16491085, 20554659).
