Pathogenic for Hereditary nonpolyposis colon cancer — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.12:g.(?_37008804)_(37012109_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 6-8 of the MLH1 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. This variant has been observed in an individual with clinical features of Lynch syndrome (PMID: 26248088). Loss-of-function variants in MLH1 are known to be pathogenic (PMID: 15713769, 24362816). For these reasons, this variant has been classified as Pathogenic.
