Pathogenic for Tuberous sclerosis 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000009.12:g.(?_132944543)_(132945143_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Studies have shown that disruption of the promoter region alters TSC1 gene expression (PMID: 14597398). A similar copy number variant has been observed in individuals with clinical features of tuberous sclerosis complex (PMID: 17287951, 20877415, 27406250; Invitae). It has also been observed to segregate with disease in related individuals. This variant is a gross deletion that occurs in a non-coding region of the TSC1 gene. It does not change the encoded amino acid sequence of the TSC1 protein.
