Pathogenic for Cystinuria — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000002.12:g.(?_44299971)_(44318104_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an in-frame deletion of the genomic region encompassing exons 5-9 of the SLC3A1 gene. It preserves the integrity of the reading frame. This variant has not been reported in the literature in individuals with SLC3A1-related conditions. Sub-genic deletion of 6-7 has been determined to be pathogenic (PMID: 28646536). Therefore, deletions that fully encompass that region are also expected to be pathogenic. This variant disrupts the p.Met467 amino acid residue in SLC3A1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 8054986, 21677404, 8792820, 23532419). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing.
