Likely pathogenic for Familial cancer of breast — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000016.10:g.(?_23621352)_(23621488_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an in-frame deletion of the genomic region encompassing exon 10 of the PALB2 gene. It preserves the integrity of the reading frame. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. This variant disrupts a portion of the WD40 domain of the PALB2 protein (residues 1000-1038), which may be essential for the interaction with the BRCA2 protein (interaction residues 1010-1109) (PMID: 19609323, 19423707, 30638972). While functional studies have not been performed to directly test the effects of this variant on PALB2 protein function,¬†this suggests that disruption of this region of the protein may abolish its DNA repair function. This variant has not been reported in the literature in individuals with PALB2-related conditions.
