Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000013.11:g.(?_32394669)_(32398770_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts the C-terminus of the BRCA2 protein. Other variant(s) that disrupt this region (p.Tyr3308*) have been determined to be pathogenic (PMID: 17026620, 18593900, 18607349, 22711857). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. A similar deletion of exons 25-27 has been observed in an individual undergoing testing for hereditary breast and ovarian cancer (PMID: 26271414). This variant is a gross deletion of the genomic region encompassing exons 25-27 of the BRCA2 gene. The 5' boundary is likely confined to intron 24. The 3' end of this event is unknown as it extends through the termination codon beyond the assayed region for this gene and may encompass additional genes. While this deletion is not anticipated to result in nonsense mediated decay, it is expected to create a truncated protein product or disrupt mRNA translation.
