Pathogenic for Phenylketonuria — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000012.12:g.(?_102894725)_(102894931_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exon 3 of the PAH gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Similar deletions of PAH exon 3 have been reported in several individuals affected with phenylketonuria (PMID: 16931086, 16931086, 17221866, 23357515, 24350308, 27175306). Loss-of-function variants in PAH are known to be pathogenic (PMID: 1301187, 9634518). For these reasons, this variant has been classified as Pathogenic.
