Pathogenic for Juvenile polyposis syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000018.10:g.(?_51029921)_(51078477_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the SMAD4 gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. Entire gene deletions of SMAD4 have been observed in several individuals affected with juvenile polyposis syndrome (PMID: 17873119). Loss-of-function variants in SMAD4 are known to be pathogenic (PMID: 16152648, 16436638, 22810475). For these reasons, this variant has been classified as Pathogenic.
