Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.11:g.(?_215837981)_(215846043_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 45-47 of the USH2A gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. A similar deletion of exons 45-47 has been reported as homozygous in an individual affected with hearing impairment (PMID: 28000701). This variant is also known as c.(8845+1_8846-1)_(9371+1_9372-1)del in the literature. Loss-of-function variants in USH2A are known to be pathogenic (PMID: 10729113, 10909849, 20507924, 25649381).
