Pathogenic for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.12:g.(?_37006981)_(37007073_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exon 5 of the MLH1 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Deletions of exon 5 have been observed in individual(s) with Lynch syndrome (PMID: 19419416, 12624159). For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in MLH1 are known to be pathogenic (PMID: 15713769, 24362816).
