Pathogenic for Glycogen storage disease, type IV; Glycogen storage disease IV, classic hepatic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.12:g.(?_81642771)_(81643000_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 7 of the GBE1 gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame. A similar copy number variant has been observed in individual(s) with glycogen storage disease type IV (PMID: 21917543). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant disrupts the p.Tyr329 amino acid residue in GBE1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 8613547, 20655781, 24082139, 25133958, 26199317). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
