Pathogenic for Multiple sulfatase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.12:g.(?_4452866)_(4467255_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 1-2 of the SUMF1 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 2 of the SUMF1 gene. This is expected to result in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with SUMF1-related conditions. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in SUMF1 are known to be pathogenic (PMID: 12757705, 12757706, 25885655).
