This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. Similar deletions of exon 4 have been observed to be homozygous or on the opposite chromosome (in trans) from other pathogenic variants in individuals and families affected with early-onset Parkinson s disease (PMID: 9560156, 9851438, 10824074, 10939576, 15254940, 21993715, 27182553), and have also been observed to segregate with disease in affected families (PMID: 9560156, 10939576, 15254940, 21993715). This finding is consistent with autosomal recessive inheritance, and suggests that this variant contributes to disease.
