Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000002.12:g.(?_227303028)_(227305093_?)dup, citing Invitae Variant Classification Sherloc (09022015): This variant results in a copy number gain of the genomic region encompassing exon(s) 44-47 of the COL4A3 gene. While the exact position of this variant cannot be determined from the data, sub-genic copy number gains are generally in tandem (PMID: 25640679). This variant is predicted to be out-of-frame, and may result in an absent or disrupted protein product. Loss-of-function variants in COL4A3 are known to be pathogenic (PMID: 8956999, 24854265, 26809805, 27281700). A similar copy number variant has been observed in individuals with Alport syndrome (PMID: 24854265, 32359821). For these reasons, this variant has been classified as Pathogenic.
