Loss-of-function variants in SMARCB1 are known to be pathogenic (PMID: 10521299, 21208904). This variant has been observed in individuals affected with schwannomas and malignant rhabdoid tumor (PMID: 24123847, 21108436). It has also been observed to segregate with disease in a family (PMID: 19124645). This variant results in a copy number gain of the genomic region encompassing exon 6 of the SMARCB1 gene. While the exact position of this variant cannot be determined from this data, sub-genic copy number gains are generally in tandem (PMID: 25640679) and may result in an absent or disrupted protein product.
