Pathogenic for ALG6-congenital disorder of glycosylation 1C — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000001.11:g.(?_63428723)_(63429136_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 13-14 of the ALG6 gene. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to disrupt the C-terminus of the protein. This variant has not been reported in the literature in individuals affected with ALG6-related conditions. This variant disrupts a region of the ALG6 protein in which other variant(s) (p.Val447Serfs*44) have been determined to be pathogenic (PMID: 23430515, 27287710; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
