NC_000022.11:g.(?_28687897)_(28725377_?)del was classified as Pathogenic for Familial cancer of breast by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 3-15 of the CHEK2 gene, which includes the termination codon. This deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. While this deletion is not anticipated to lead to nonsense mediated decay, it is expected to alter mRNA translation or result in a truncated protein product. This variant has not been reported in the literature in individuals affected with CHEK2-related conditions. This variant disrupts a region of the CHEK2 protein in which other variant(s) (exon 3-4 deletion) have been determined to be pathogenic (PMID: 18004398, 24763289). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
