Pathogenic for Retinoblastoma — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000013.11:g.(?_48303701)_(48304059_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon 1 of the RB1 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 1 of the RB1 gene. This is expected to result in an absent or disrupted protein product. Similar deletion of exon 1 has been observed in individual(s) with retinoblastoma (PMID: 22180099). This variant is also described as c.1_137del in the literature. Loss-of-function variants in RB1 are known to be pathogenic (PMID: 17096365). For these reasons, this variant has been classified as Pathogenic.
