Pathogenic for Carney complex, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.11:g.(?_68512379)_(68515586_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 1-2 of the PRKAR1A gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 2 of the PRKAR1A gene. This is expected to result in an absent or disrupted protein product. Smaller deletions of exon 2 have been observed in individual(s) with clinical features of Carney complex (PMID: 28640241). Loss-of-function variants in PRKAR1A are known to be pathogenic (PMID: 11115848, 19293268). For these reasons, this variant has been classified as Pathogenic.
