Pathogenic for Hypohidrotic X-linked ectodermal dysplasia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.11:g.(?_69957007)_(69957152_?)dup, citing Invitae Variant Classification Sherloc (09022015): This variant results in a copy number gain of the genomic region encompassing exon(s) 2 of the EDA gene. While the exact position of this variant cannot be determined from the data, sub-genic copy number gains are generally in tandem (PMID: 25640679). This variant is predicted to be out-of-frame, and may result in an absent or disrupted protein product. Loss-of-function variants in EDA are known to be pathogenic (PMID: 9683615). A similar copy number variant has been observed in individual(s) with X-linked hypohidrotic ectodermal dysplasia (PMID: 24689965, 27305980). For these reasons, this variant has been classified as Pathogenic.
