This variant lies in the PALB2 gene (transcript NM_024675.4) at coding-DNA position 1910, where C is replaced by A; at the protein level this means replaces proline at residue 637 with histidine — a missense variant. Submitter rationale: The p.P637H variant (also known as c.1910C>A), located in coding exon 5 of the PALB2 gene, results from a C to A substitution at nucleotide position 1910. The proline at codon 637 is replaced by histidine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6496 samples (12992 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.001% (greater than 130000 alleles tested) in our clinical cohort. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence for this variant is limited at this time, the clinical significance of p.P637H remains unclear.
