Uncertain significance for Long QT syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033337.3(CAV3):c.86C>T (p.Pro29Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 29 of the CAV3 protein (p.Pro29Leu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of autosomal dominant caveolinopathies and/or hyperCKemia (PMID: 12082049; Invitae). This variant is also known as p.Pro28Leu. ClinVar contains an entry for this variant (Variation ID: 8287). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects CAV3 function (PMID: 14749532, 20472890).
