This variant lies in the FZD6 gene (transcript NM_003506.4) at coding-DNA position 1525, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 509 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The variant c.1525C>T (p.Arg509Ter) in the FZD6 gene causes a nonsyndromic congenital nail disorder, which has previously been reported in families with an autosomal recessive mode of inheritance. In those families, the variant segregated with the disease, including both affected and unaffected members. According to our study, we observed some novel phenotypes involving the teeth, skin, and hair, in addition to the nail abnormalities with autosomal recessive mode of inheritance pattern. and segregated in all the individuals affected and unaffected.
