NM_000535.7(PMS2):c.829A>C (p.Thr277Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T277P variant (also known as c.829A>C), located in coding exon 8 of the PMS2 gene, results from an A to C substitution at nucleotide position 829. The threonine at codon 277 is replaced by proline, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

Protein context (NP_000526.2, residues 267-287): FYISGFISQC[Thr277Pro]HGVGRSSTDR
