NM_000051.4(ATM):c.8110T>C (p.Cys2704Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C2704R variant (also known as c.8110T>C), located in coding exon 54 of the ATM gene, results from a T to C substitution at nucleotide position 8110. The cysteine at codon 2704 is replaced by arginine, an amino acid with highly dissimilar properties. This alteration was previously identified in a Portuguese patient with ataxia telangiectasia (Demuth I et al. Neurogenetics, 2011 Nov;12:273-82). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
