This variant lies in the PRKAR1A gene (transcript NM_002734.5) at coding-DNA position 791, where G is replaced by A; at the protein level this means replaces arginine at residue 264 with histidine — a missense variant. Submitter rationale: Variant summary: PRKAR1A c.791G>A (p.Arg264His) results in a non-conservative amino acid change located in the cyclic nucleotide-binding domain (IPR000595) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251324 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.791G>A in individuals affected with Carney Complex and no experimental evidence demonstrating its impact on protein function have been reported.
