Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005591.4(MRE11):c.748A>G (p.Lys250Glu), citing Ambry Variant Classification Scheme 2023: The p.K250E variant (also known as c.748A>G), located in coding exon 7 of the MRE11A gene, results from an A to G substitution at nucleotide position 748. The lysine at codon 250 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
