NM_024642.5(GALNT12):c.744G>C (p.Glu248Asp) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E248D variant (also known as c.744G>C), located in coding exon 4 of the GALNT12 gene, results from a G to C substitution at nucleotide position 744. The glutamic acid at codon 248 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
