Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.7360G>C (p.Ala2454Pro), citing Ambry Variant Classification Scheme 2023: The p.A2454P variant (also known as c.7360G>C), located in coding exon 49 of the ATM gene, results from a G to C substitution at nucleotide position 7360. The alanine at codon 2454 is replaced by proline, an amino acid with highly similar properties. In an assay testing ATM function, this variant showed a functionally abnormal result. (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
