NM_000455.5(STK11):c.65T>G (p.Met22Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M22R variant (also known as c.65T>G), located in coding exon 1 of the STK11 gene, results from a T to G substitution at nucleotide position 65. The methionine at codon 22 is replaced by arginine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
