Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000075.4(CDK4):c.632A>T (p.Lys211Met), citing Ambry Variant Classification Scheme 2023: The p.K211M variant (also known as c.632A>T), located in coding exon 4 of the CDK4 gene, results from an A to T substitution at nucleotide position 632. The amino acid change results in lysine to methionine at codon 211, an amino acid with similar properties. However, this change occurs in the last base pair of coding exon 4, which makes it likely to have some effect on normal mRNA splicing. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
