Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.6303A>C (p.Gln2101His), citing Ambry Variant Classification Scheme 2023: The p.Q2101H variant (also known as c.6303A>C), located in coding exon 42 of the ATM gene, results from an A to C substitution at nucleotide position 6303. The glutamine at codon 2101 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
