NM_006361.6(HOXB13):c.601+1G>T was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.601+1G>T intronic variant results from a G to T substitution one nucleotide after coding exon 1 of the HOXB13 gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, loss of function of HOXB13 has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this alteration remains unclear.
