Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002485.5(NBN):c.589dup (p.Tyr197fs), citing Ambry Variant Classification Scheme 2023: The c.589dupT pathogenic mutation, located in coding exon 6 of the NBN gene, results from a duplication of T at nucleotide position 589, causing a translational frameshift with a predicted alternate stop codon (p.Y197Lfs*5). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
