NM_000314.8(PTEN):c.586C>G (p.His196Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H196D variant (also known as c.586C>G), located in coding exon 6 of the PTEN gene, results from a C to G substitution at nucleotide position 586. The histidine at codon 196 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
