Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.5837T>G (p.Leu1946Trp), citing Ambry Variant Classification Scheme 2023: The p.L1925W variant (also known as c.5774T>G), located in coding exon 39 of the NF1 gene, results from a T to G substitution at nucleotide position 5774. The leucine at codon 1925 is replaced by tryptophan, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
