NM_177438.3(DICER1):c.5422A>C (p.Met1808Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M1808L variant (also known as c.5422A>C), located in coding exon 24 of the DICER1 gene, results from an A to C substitution at nucleotide position 5422. The methionine at codon 1808 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
