Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.5119A>C (p.Ile1707Leu), citing Ambry Variant Classification Scheme 2023: The p.I1707L variant (also known as c.5119A>C), located in coding exon 16 of the BRCA1 gene, results from an A to C substitution at nucleotide position 5119. The isoleucine at codon 1707 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
