Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.4932G>A (p.Met1644Ile), citing Ambry Variant Classification Scheme 2023: The p.M1644I variant (also known as c.4932G>A), located in coding exon 32 of the ATM gene, results from a G to A substitution at nucleotide position 4932. The methionine at codon 1644 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
