Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.4811A>G (p.Asp1604Gly), citing Ambry Variant Classification Scheme 2023: The p.D1604G variant (also known as c.4811A>G), located in coding exon 31 of the ATM gene, results from an A to G substitution at nucleotide position 4811. The aspartic acid at codon 1604 is replaced by glycine, an amino acid with similar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
