NM_001042492.3(NF1):c.480G>T (p.Arg160Ser) was classified as Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R160S variant (also known as c.480G>T) is located in coding exon 5 of the NF1 gene. The arginine at codon 160 is replaced by serine, an amino acid with dissimilar properties. This change occurs in the first base pair of coding exon 5. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
