Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.4622A>G (p.Lys1541Arg), citing Ambry Variant Classification Scheme 2023: The p.K1541R variant (also known as c.4622A>G), located in coding exon 22 of the DICER1 gene, results from an A to G substitution at nucleotide position 4622. The lysine at codon 1541 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
