NM_005732.4(RAD50):c.443A>T (p.Lys148Met) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K148M variant (also known as c.443A>T), located in coding exon 4 of the RAD50 gene, results from an A to T substitution at nucleotide position 443. The lysine at codon 148 is replaced by methionine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
